琼脂糖凝胶电泳所用缓冲液的类型可由DNA片段的大小和迁移后实验的含量来决定。
如果你想分离大于或等于15,000 bp的大DNA，使用TAE缓冲液。如果你不想提取DNA，或者不关心迁移后1000bp或更小的DNA的恢复率，可以使用TBE缓冲液。
上图显示了EzDNA Ladder应用于使用EzRunTAE或EzRunTBE缓冲液的1.5%琼脂糖凝胶，并使用dive – mini在150V下电泳20分钟的结果。可以看到，TAE缓冲液适用于聚合物分离，TBE缓冲液适用于低分子分离。
